Mam actually inserting the dna means Only small piece of dna is used and how it alters the existing Dna.. The existing DNA is stable right, then where does it go and bind??? and also only one cell is been combined with rdna then how the entire progency will get the character change due to change in one r 2 cells..i.e. where it is done exactly in host organisms?? By gel electrophoresis u said we can identify our gene of interest how by looking we can understand the gene of interest by looking we dont know the sequence and by looking we cant say it is my gene of interest?? Pls clarify...
Answer:

Inserting only small piece of DNA would alter the existing DNA as that small piece will make a change in genes and due to which the genetic expression also changes. By combining with only one DNA makes the change in existing gene expression. 

By using gel electrophoresis, we can identify gene of interest as the fragments of DNA have different sizes. Sequences of DNA is different for every species. We separate fragments by using gel electrophoresis and amplify them using polymerase chain reactions.
