Question:
How can do we insert the DNA in the cell, while the DNA is stable in the host nucleus.. actually we have the parts of DNA for eye color,etc.. exactly how do we find the place where the gene responsible for our interest is located... and another question is inserting rDNA in one cell how can it change the genes which are to go for sexual reproduction.Actually only one cell in body is changed,how can it infect the sex cells which have genes to pass for progency i.e., how can it express in progeny also as there is only 1-5 somatic cell changed by rDNA?
Answer:

If we have part of some DNA, we will ligate that DNA fragment on plasmid vector and then transfer it into host cell say E.Coli. It can be done by gene gun also.

And the way we can find out the gene of interest is located by complimentary restriction endonuclease enzyme. These enzymes can cut DNA only on a specific site, this is due to the DNA sequence of that gene.

rDNA refers to the recombinant DNA, the cell carrying rDNA is highly reproductive in nature so they start replicating in very large number and replace all cells in a body because cells are not immortal, they are replaced by new DNA. The germcell in spermatogonia , in males, get affected by rDNA only if they possess the capability to change it.

As you know gametogenesis also arise from cell differentiation.
